How do we use the straddling command in expression gui of the rectangle command in Pcell designer tool. What purpose does the reference and test shapes serve in here.

    The idea is that the straddling operator finds test shapes which are partly covered by the reference shape, and partly not covered by the reference shape (in other words, the test shape straddles the boundary of the reference shape). Something like this:
